Product Overview

Key Features

Ambrisentan is a prescription medication used to treat pulmonary arterial hypertension (PAH), a serious condition characterized by high blood pressure in the arteries that carry blood from the heart to the lungs. By lowering this pressure, it makes it easier for the heart to pump blood, improves a person's ability to exercise, and helps to delay the worsening of the condition. Active Ingredient Generic Name: Ambrisentan Chemical Class: Endothelin Receptor Antagonist (ERA). Specifically, it is a selective antagonist of the endothelin type A (ETA) receptor. Mechanism of Action: In individuals with PAH, the levels of a hormone called endothelin are abnormally high. Endothelin causes the blood vessels in the lungs to constrict and their walls to thicken. Ambrisentan works by blocking the ETA receptors, which are responsible for these harmful effects. By doing so, it helps the blood vessels in the lungs to relax and widen, reducing the blood pressure and the strain on the heart. Indications (Uses) Pulmonary Arterial Hypertension (PAH): Ambrisentan is prescribed for adults with PAH (WHO Group 1) to improve their exercise ability and delay clinical worsening. It may be used as a single agent or in combination with other medications like tada-lafil to further reduce the risk of disease progression. Dosage and Administration Forms: Ambrisentan is available as oral tablets. Tablet Strengths: Common tablet strengths are 5 mg and 10 mg. Typical Dosing: The initial dose for adults is typically 5 mg once a day. A doctor may increase the dose to 10 mg once a day based on how the patient tolerates the medication and their clinical response. Food: It can be taken with or without food. Administration: Tablets should be swallowed whole and not crushed, chewed, or broken. Common Brand Names Letairis Volibris Important Precautions and Side Effects Liver Function: Ambrisentan can cause an increase in liver enzyme levels. Regular blood tests are necessary to monitor liver function, particularly at the start of treatment. Pregnancy and Birth Control: Ambrisentan is extremely dangerous for unborn babies and can cause serious birth defects. Women who are able to become pregnant must have a negative pregnancy test before starting treatment, and they must use two reliable forms of birth control during treatment and for at least one month after stopping the medication. Anemia: The medication may cause a decrease in red blood cell count (anemia). Regular blood tests are typically performed to monitor for this side effect. Fluid Retention: Swelling of the hands, ankles, or feet (peripheral edema) is a common side effect. Patients should report any rapid weight gain or swelling to their doctor, as this could be a sign of fluid retention. Other Side Effects: Other common side effects include headache, dizziness, nasal congestion, and flushing. Drug Interactions: Ambrisentan can interact with other medications, particularly cyclosporine. It is crucial to inform your doctor about all prescription and over-the-counter medications, vitamins, and supplements you are taking.
