Uses

Dasatinib is atyrosine kinase inhibitor (TKI)used to treat:

• Chronic Myeloid Leukemia (CML) in chronic, accelerated, or blast phase
• Philadelphia chromosome-positive (Ph+) Acute Lymphoblastic Leukemia (ALL)
• CML or Ph+ ALLresistant or intolerant to prior therapy, including imatinib

Dosage & Administration

• Standard dose for adults: 50100mgonce daily, orally
• May vary depending on disease phase & patient factors
• Takewith or without food
• Swallow whole tablet;do not crush or split
• Dose adjustments required foradverse effectsorlow blood counts

Common Side Effects

• Low blood counts: neutropenia, thrombocytopenia, anemia
• Fluid retention: pleural effusion, edema
• Diarrhea, nausea, fatigue
• Headache, rash
• Muscle or joint pain
• Cough, shortness of breath

Monitoring

• CBC (Complete Blood Count): weekly in initial phase, then monthly
• Liver & kidney function tests
• Chest X-ray or ECGif shortness of breath or fluid retention occurs
• Electrolytes& cardiac monitoring in at-risk patients

Precautions

• Avoid use inpregnancy (Category D) may harm fetus
• Caution in patients withlung or heart conditions
• Can causeQT prolongation use carefully with heart medications
• Not recommended insevere hepatic impairment
• Avoidgrapefruit juice may increase toxicity
• May suppress bone marrow monitor blood counts closely

Drug Interactions

• CYP3A4 inhibitors(e.g., ketoconazole) dasatinib levels
• CYP3A4 inducers(e.g., rifampin) dasatinib levels
• Antacids or PPIs reduce dasatinib absorption (separate by several hours)
• Caution withQT-prolonging drugs

Tips for Patients

• Take at the same time each day
• Report signs ofinfection, bleeding, or breathlessness immediately
• Avoid over-the-counteracid-reducing agents
• Use effectivecontraceptionduring & after treatment
