Thyroid-stimulating hormone (TSH or thyrotropin) is a glycoprotein hormone synthesized and secreted by the anteriorlobe of
the pituitary gland, which regulates the endocrine function of the thyroid.Structurally, the glycoprotein hormones are related
heterodimers comprised of a common alpha subunit and a hormone-specific beta subunit, which are non-covalently bound to
one another. The alpha subunit of TSH is identical to that ofhuman chorionic gonadotropin (HCG), luteinising hormone (LH) and
follicle-stimulating hormone (FSH). The beta subunitis TSH-specific, and therefore determines its function.TSH production is
controlled by hypothalmic-derived thyrotropin releasing hormone (TRH). Circulating TSH in the blood isthe primary determining
factor that regulates the secretion of the thyroid hormones thyroxine (T4) and tri-iodothyronine(T3) from the thyroid. The level of
thyroid hormones in the blood has an effect on the pituitary release of TSH; when thelevels of T3 and T4 are low, the production
of TSH is increased, and conversely, when levels of T3 and T4 are high, TSHproduction is decreased. This effect creates a
regulatory negative feedback loop.
