Triglyceride in the sample is hydrolyzed by microbial lipases to glycerol and free fatty acid (FFA). Glycerol is phosphorylated by ATP to glycerol -3-phosphate (G-3-P) in reaction catalyzed by G.K. G-3-P is oxidized to D.A.P. in a reaction catalyzed by the enzyme GPO. In this reaction H2 O2 is produced in equimoler concentration to the level of triglycerides present in the sample. H2 O2 with 4-AAP and DHBS in reaction in a reaction catalyzed by HPOD. The result of this oxidative coupling is a quinoneimine red complex dye. The absorbance of this dye in solution is proportional to the concentration of triglyceride in the sample.
