It is applied directly into the nose, either as a spray or as drops. Mechanism of action: Xylometazoline is a sympathomimetic agent with marked alpha-adrenergic activity, and is intended for intranasal use. It is a sympathomimetic amine of the imidazoline class. It act directly on alpha-adrenoreceptors. But does not act on beta-receptors. It constricts the nasal blood vessels, thereby decongesting the
mucosa of the nose andneighbouring regions of the pharynx. This enables patients suffering from colds tobreathe more easily through the nose. Xylometazoline is generally well tolerated anddoes not impair the function of the ciliated epithelium. Onset of action is within 5-10minutes, with the decongestant effect being prolonged and lasting for up to 10 hours. Xylometazoline was found to have a high affinity for alpha-adrenergic receptors. Stimulation of the alpha-receptors induces a signalling cascade, which ends in an
increase in the intracellular calcium concentration. Calcium stimulates myosinkinase, an enzyme that activates the myosin filament of the muscles.
