1. Autoclave

An Autoclave is a device which is used to sterilize and supplies and equipment by exposing them to high pressure saturated steam at the temperature level of 121 C or more, ideally for 15-20 minutes.

In a layman term, we can say that autoclave is a pressure cooker. Water is heated up in a pressurized container to generate steam. By the use of pressure water is heated to a higher level without the consumption of extra energy. Usually, steel is utilized for making Autoclaves. Their are different configurations for withdrawing air before the stage of pressurization. Gravity is used by the downward displacement autoclaves to remove air. Steam pulsing autoclaves have a mechanism of pressurizing and depressurizing by the use of pulses of steam to attain optimum pressure. Vacuum pump autoclaves throw out air pressurization. Superatmospheric autoclaves are the accumulation of vacuum pump and steam pulsing techniques. Autoclaves are widely used in medicine, microbiology, body piercing, tattooing, mycology, veterinary science, chiropody, dentistry and prosthetic fabrication.

Typical loads comprise surgical instruments, laboratory glassware, patient care utensils, medical waste, Lysogeny broth and animal cage bedding.

A popularly growing application of autoclaves is in the sterilization and pre-disposal treatment of waste materials, like pathogenic hospital waste. In this category, machines largely function under the identical principles as of the original autoclave. Hence, they are able to neutralize potentially infectious agents by implementing superheated water and pressurized steam.

How Does an Autoclave Work?

An autoclave disinfects items by heating them with the help of steam to a remrkably high temperature. Some common operational temperature level for autoclaves are: 121 degrees C/15 p.s.i., 115 degrees C/10 p.s.i., and 134 degrees C/30 p.s.i. (p.s.i.=pounds per square inch). The temperature, time of operation and pressure depend on the level of sterilization needed.

Autoclave Quality Assurance

Bags/pads for Sterilization often have a "sterilization indicator mark" that ideally darkens as the bag/pad is processed. Comparison between the unprocessed bag mark to a well processed bag will demonstrate a broad visual difference. There are chemical, physical and biological indicators that could be followed to assure an autoclave attains the correct temperature for the correct measure of time.

Autoclave tape and medical packaging have chemical indicators which change color once the accurate conditions have been met. This change in color indicates that the object under the tape or inside the package, has been well processed. Some computer-assisted autoclaves utilize an F0 (F-nought) value to control the sterilization cycle. Setting parameters of F0 values are number of minutes of equivalent sterilization at 121 C (250 F) at 15 psi (100 kPa) higher than the atmospheric pressure, for 15 minutes. As it is difficult to control the exact temperature, the sterilization time is adjusted after monitoring the temperature.
