An ocular micrometer is a glass disc with a ruled scale that is designed to fit in the eyepiece of a microscope and is used for the purpose of measuring the size of objects that have been magnified. The degree of magnification affects the length of the markings on the scale in terms of their actual physical dimensions.
When employing the scanning lens, low-power lens, high-power lens, or oil immersion lens, the full ocular scale may be broken down into one hundred smaller units that can be seen by the user. When using a certain objective lens, it is essential to have a solid understanding of the actual length of each ocular unit, which is measured in micrometers (m). In order to compensate for the magnification, the specimen measurements will be multiplied by a unique number that corresponds to each objective lens. When scientists calibrate the ocular scales on their microscopes, it doesn't matter what kind of microscope they use; they may still communicate with one another about the actual sizes of the objects.
An ocular micrometre acts as a scale or rule when it comes to measuring an item that is seen via a microscope. This is only a glass disc that has been engraved with divisions at equal intervals throughout its surface. The regulation may be broken down into as many as 100 subsections, but this occurs far less often. In order to make use of the ocular micrometre, you will first need to calibrate it against a recognised and stable ruler, which is the stage micrometre. Here are the steps to compute ocular and stage micrometers: 

  • Put a stage micrometre on the stage of the microscope, and then concentrate your observation on the grid that's on the stage micrometre by reducing the magnification to 4 times.

  • You may rotate the ocular micrometre by rotating the eyepiece that corresponds to it. After moving the stage to the point where you can superimpose the lines of the ocular micrometre onto those of the stage micrometre, stop. After ensuring that the lines of both micrometres are aligned at one side of the field, proceed to count the spaces between each micrometre until you reach a point where the lines of both micrometres are aligned once again.

  • Since every division of the stage, micrometre is 10 micrometres in size, and since you already know how many ocular divisions are equal to one stage division, you are now able to compute the number of micrometres that are contained inside each area on the ocular scale.

  • Repeat this process ten times, then forty times, and finally one hundred times, and record your results.
